Determination of biological paternity

Main “architect” of every organism is genetic information that is encoded in cell DNA molecules. Formal resemblance between two persons is often testifies their relatedness, however, subjective evaluation of external features is insufficient for genetic identification of person and for the determination of paternity / maternity.

Child’s biologic relation with his parent is the inheritance of gene patterns of his parents (children inherit 50% of mother genetic material and 50% of father genetic material). Director of one of the German genetic centers described determination of biological paternity in a following “colorful” way: “Imagine that parents have necklaces of different colors (mother has red, and father has blue necklaces), and they decide to present these necklaces to their children. They thread collar from their red and blue beads. If you imagine that these collar beads are genes, you will clearly see that child has only “red” and “blue” genes. There are no beads (genes) of other colors in child's collar”.

To identify person DNA are usually separated from blood and cells of oral cavity epithelium. Human DNA could also be separated from bones, hair, sperm, nails and other tissues.

In the laboratory of the SOPRO medical research center human DNA is separated from blood and cells of oral epithelium. This method is painless and non-invasive, and therefore in the recent years determination of biological paternity by obtaining DNA from cells of oral epithelium became the most popular globally.

Biological paternity test is performed using modern technique of molecular biology, PCR method, and kits of reagents of highest quality are used. In the course of reaction 15 specific DNR regions, so called DNA markers, are replicated. Specific DNA markers are short tandem polymorphic nucleotide sequences, STRs (Short Tandem Repeats) forming many hundred thousand copies in the human genome. Reaction products are analyzed by automatic analyzer.

After analysis of the DNA genetic profiles are composed for father and child or for father, child and mother; and these genetic profiles comprise unique sets of fifteen specific DNA markers (usually examination of child’s mother is not necessary for the identification of biological paternity; however in some cases it could enhance accuracy of the test). Comparison of genetic profiles ensures accurate and reliable determination of biological paternity (maternity) of examined persons.

Presently accuracy of the determination of biological paternity using molecular genetic technique reaches 99,999…%.

All persons under investigation are obliged to take part in sampling. All personal data are confidential (anonymous test is possible). Only coded samples access the laboratory. Findings are not presented or commented by phone. Findings are issued in the SOPRO laboratory or are sending by e-mail or by registered letter.

Venous (capillary) blood or cells of oral epithelium are used for the test. Blood and oral epithelium cell specimens are obtained in SOPRO laboratory every working day and on Saturdays. Test results are presented after 2-3 weeks.
